Other missense substitutions at this codon (p.Thr55Ile, p.Thr55Arg, and p.Thr55Ala) have been reported in individuals affected with Charcot-Marie-Tooth disease  (PMID: 10220155, 12185164, 12477701, 11271367). Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with GJB1-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces threonine with proline at codon 55 of the GJB1 protein (p.Thr55Pro). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and proline. - |
