However, these predictions have yet to be confirmed by functional studies. The variant was absent in 31396 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.316-189A>G in individuals affected with Hemoglobinopathy and no experimental evidence demonstrating its impact on protein function have been reported. However, other variants within intron 2 have been previously classified as pathogenic (e.g. c.316-3C>A, c.316-7C>A, c.316-14T>G, c.316-106C>G, c.316-146T>G, c.316-197C>T, and c.316-238C>T). Two other clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. One laboratory classified the variant as likely benign, and one laboratory classified the variant as uncertain significance.
