Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 251478 control chromosomes. c.947T>A has been reported in the literature in individuals affected with Leukoencephalopathy With Vanishing White Matter, in compound heterozygosity with pathogenic variants (Leegwater_2001, Slynko_2020). These data indicate that the variant is likely to be associated with disease. Experimental studies reported the vairant to have significantly reduced eIF2B activity (Richardson_2004) and partial loss of activity with reduced efficienciency to form holocomplexes (Li_2004). One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation.
