The glycine at codon 426 is replaced by serine, an amino acid with similar properties. This alteration was reported in 1/469 cases with multiple colorectal adenomas and/or familial CRC; however, structural studies showed that this alteration is unlikely to be involved in direct DNA binding, and was predicted to have only moderate functional effects (Palles C et al. Nat. Genet. 2013 Feb;45(2):136-44). This alteration was also identified in an individual diagnosed with colorectal cancer (Chubb D et al. J Clin Oncol, 2015 Feb;33:426-32).This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
