A different variant affecting this nucleotide has been reported in a patient affected with nevoid basal cell carcinoma syndrome (PMID: 8981943), indicating that this nucleotide may be crucial for normal RNA splicing. This variant has not been reported in the literature in individuals with a PTCH1-related disease. However, it has been observed to segregate with disease in a single family (Invitae). This sequence change affects an acceptor splice site in intron 7 of the PTCH1 gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. -
