While this particular variant has not been reported in the literature, loss-of-function variants in BMPR1A are known to be pathogenic (PMID: 11536076, 12417513). This sequence change deletes 4 nucleotides from exon 10 of the BMPR1A mRNA (c.949_952delCTCT), causing a frameshift at codon 317. This creates a premature translational stop signal (p.Leu317Metfs*4) and is expected to result in an absent or disrupted protein product. -
