Genes affected
BMPR1A (HGNC:1076): (bone morphogenetic protein receptor type 1A) The bone morphogenetic protein (BMP) receptors are a family of transmembrane serine/threonine kinases that include the type I receptors BMPR1A and BMPR1B and the type II receptor BMPR2. These receptors are also closely related to the activin receptors, ACVR1 and ACVR2. The ligands of these receptors are members of the TGF-beta superfamily. TGF-betas and activins transduce their signals through the formation of heteromeric complexes with 2 different types of serine (threonine) kinase receptors: type I receptors of about 50-55 kD and type II receptors of about 70-80 kD. Type II receptors bind ligands in the absence of type I receptors, but they require their respective type I receptors for signaling, whereas type I receptors require their respective type II receptors for ligand binding. For these reasons, this variant has been classified as Pathogenic. While this particular variant has not been reported in the literature, loss-of-function variants in BMPR1A are known to be pathogenic (PMID: 11536076, 12417513). This sequence change deletes 4 nucleotides from exon 10 of the BMPR1A mRNA (c.949_952delCTCT), causing a frameshift at codon 317. This creates a premature translational stop signal (p.Leu317Metfs*4) and is expected to result in an absent or disrupted protein product. -

The c.949_952delCTCT pathogenic mutation, located in coding exon 8 of the BMPR1A gene, results from a deletion of 4 nucleotides at nucleotide positions 949 to 952, causing a translational frameshift with a predicted alternate stop codon (p.L317Mfs*4). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
