The variant allele was found at a frequency of 2.4e-05 in 250998 control chromosomes. c.1606T>C has been reported in the literature in the homozygous state in multiple individuals affected with Usher Syndrome (example, Dad_2016, Neuhaus_2017). These data indicate that the variant is likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in completely abolished binding activity in vitro to partner protein, collagen IV (example, Bhattacharya_2004).
