It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 48471). Variant summary: USH2A c.1606T>C (p.Cys536Arg) results in a non-conservative amino acid change located in the Laminin-type EGF domain (IPR002049) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.4e-05 in 250998 control chromosomes. c.1606T>C has been reported in the literature in the homozygous state in multiple individuals affected with Usher Syndrome (example, Dad_2016, Neuhaus_2017). These data indicate that the variant is likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in completely abolished binding activity in vitro to partner protein, collagen IV (example, Bhattacharya_2004). The following publications have been ascertained in the context of this evaluation (PMID: 14676276, 27957503, 28944237). ClinVar contains an entry for this variant (Variation ID: 48471).
