This sequence change creates a premature translational stop signal (p.Arg737*) in the USH2A gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in USH2A are known to be pathogenic (PMID: 10729113, 10909849, 20507924, 25649381). This variant is present in population databases (rs111033334, gnomAD 0.003%). This premature translational stop signal has been observed in individual(s) with Usher syndrome and autosomal recessive retinitis pigmentosa (PMID: 17296898, 25356976, 29490346). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 2361). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. -
