The c.1529_1530delCA pathogenic mutation, located in coding exon 9 of the MEN1 gene, results from a deletion of two nucleotides between positions 1529 and 1530, causing a translational frameshift with a predicted alternate stop codon. This mutation has been reported in one German family whose history included primary hyperparathyroidism, prolactinoma, insulinoma, and carcinoid of the thymus. The authors referred to this alteration as 510fs19aaX (Bartsch D et al. Eur. J. Endocrinol. 1998 Oct;139(4):416-20). In addition to the clinical data presented in the literature, since frameshifts are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294). -
