For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 428709). This variant is also known as c.2463+1G>A. Disruption of this splice site has been observed in individual(s) with retinoblastoma (PMID: 15884040, 27582626, 28193182). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change affects a donor splice site in intron 22 of the RB1 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in RB1 are known to be pathogenic (PMID: 17096365). -

The c.2325+1G>A intronic pathogenic mutation results from a G to A substitution one nucleotide after coding exon 22 of the RB1 gene. This mutation, referred to as c.2463+1G>A, has been reported in the germline of an individual with bilateral retinoblastoma (Nichols KE et al. Hum. Mutat. 2005 Jun;25(6):566-74). It was also seen in an Indian retinoblastoma cohort (Singh J et al. Mol. Vis. 2016 Aug;22:1036-47). In addition to the clinical data presented in the literature, alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is classified as a disease-causing mutation. -
