Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251302 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2137G>C has been reported in the literature as a compound heterozygous genotype in at-least one individual affected with Trichothiodystrophy who has been subsequently cited by others (example, Takayama_1996, Taylor_1997, Botta_1998, Lanzafame_2022). These data do not allow any conclusion about variant significance. At least one publication reports experimental evidence evaluating an impact on protein function reporting abolished interaction with the p44 subunit of TFIIH resulting in reduced helicase activity of XPD (Dubaele_2003). No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014.
