This alteration results from a C to T substitution at nucleotide position 299, causing the serine (S) at amino acid position 100 to be replaced by a phenylalanine (F). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This variant was detected in an extra-adrenal pheochromocytoma, which showed loss of heterozygosity and loss of SDHB on immunochemistry (van Nederveen, 2007; Korpershoek, 2007). Another alteration at the same codon, c.298T>C (p.S100P), has been detected in multiple individuals with features consistent with SDHB-related paraganglioma-pheochromocytoma syndrome (Pollard, 2005; Yalcintepe, 2021; Güleç Ceylan, 2022). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis.
