Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with hypertrophic cardiomyopathy (PMID: 15358028) and in an individual affected with sudden cardiac arrest (PMID: 37194601). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The p.K1919N variant (also known as c.5757G>C), located in coding exon 37 of the MYH7 gene, results from a G to C substitution at nucleotide position 5757. The lysine at codon 1919 is replaced by asparagine, an amino acid with some similar properties. This alteration was reported in both cases and controls in a hypertrophic cardiomyopathy (HCM) cohort (Van Driest SL et al. J. Am. Coll. Cardiol., 2004 Aug;44:602-10). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
