GDF2 (HGNC:4217): (growth differentiation factor 2) This gene encodes a secreted ligand of the TGF-beta (transforming growth factor-beta) superfamily of proteins. Ligands of this family bind various TGF-beta receptors leading to recruitment and activation of SMAD family transcription factors that regulate gene expression. The encoded preproprotein is proteolytically processed to generate each subunit of the disulfide-linked homodimer. This protein regulates cartilage and bone development, angiogenesis and differentiation of cholinergic central nervous system neurons. Mutations in this gene are associated with hereditary hemorrhagic telangiectasia. [provided by RefSeq, Jul 2016]

This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 283 of the GDF2 protein (p.Val283Met). This variant is present in population databases (rs138904328, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with GDF2-related conditions. ClinVar contains an entry for this variant (Variation ID: 430360). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt GDF2 protein function with a negative predictive value of 80%. A variant of uncertain significance has been identified in the GDF2 gene. The V283M variant has not been published as pathogenic or been reported as benign to our knowledge. This variant has been observed in 7/66686 (90.01%) alleles from individuals of Non-Finnish European ancestry in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The V283M variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. Additionally, this substitution occurs at a position that is not conserved across species and methionine (M) is the wild-type residue at this position in multiple mammalian species. Finally, in silico analysis predicts this variant likely does not alter the protein structure/function. -
