This sequence change falls in intron 37 of the FBN1 gene. It does not directly change the encoded amino acid sequence of the FBN1 protein, but it affects a nucleotide within the consensus splice site of the intron. This variant is not present in population databases (rs140633, ExAC no frequency). This variant has been reported an individual with incomplete Marfan syndrome and an individual with ectopia lentis (PMID: 17657824, Invitae). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may alter RNA splicing, but this prediction has not been confirmed by published transcriptional studies. In summary, this variant is a rare intronic change with uncertain impact on splicing.
