This variant has also been identified in 12/251272 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Variant summary: FBN1 c.3845A>G (p.Asn1282Ser) results in a conservative amino acid change located in the EGF-like domain (IPR000742) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4.8e-05 in 251272 control chromosomes, predominantly at a frequency of 0.00013 within the South Asian subpopulation in the gnomAD database. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3845A>G has been reported in the literature in individuals affected with Marfan Syndrome, without strong evidence for causality (example, Groth_2016, Liu_1997, Yang_2014,Yoon_2023). These report(s) do not provide unequivocal conclusions about association of the variant with Marfan Syndrome.
