Proteins belonging to this family play a central role in inducible gene transcription during the immune response. This protein regulates gene expression induced by osmotic stress in mammalian cells. Unlike monomeric members of this protein family, this protein exists as a homodimer and forms stable dimers with DNA elements. This alteration results from a G to A substitution at nucleotide position 4336, causing the glycine (G) at amino acid position 1446 to be replaced by a serine (S). This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 1352 of the NFAT5 protein (p.Gly1352Ser). This variant is present in population databases (rs145862239, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with NFAT5-related conditions. ClinVar contains an entry for this variant (Variation ID: 566168). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign".
