Variant summary
Our verdict is Uncertain significance. The variant received 4 ACMG points: 4P and 0B. PP3_ModeratePP5_Moderate
The NM_002900.3(RBP3):c.3238G>A(p.Asp1080Asn) variant causes a missense change involving the alteration of a conserved nucleotide. The variant allele was found at a frequency of 0.0000062 in 1,613,838 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Pathogenic (★).

This variant is present in population databases (rs146150511, gnomAD 0.007%). This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 1080 of the RBP3 protein (p.Asp1080Asn). ClinVar contains an entry for this variant (Variation ID: 50368). For these reasons, this variant has been classified as Pathogenic. Experimental studies have shown that this missense change affects RBP3 function (PMID: 23486466). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This missense change has been observed in individual(s) with retinitis pigmentosa (PMID: 19074801). It has also been observed to segregate with disease in related individuals. -
