Genes affected
TSPEAR (HGNC:1268): (thrombospondin type laminin G domain and EAR repeats) This gene encodes a protein that contains a N-terminal thrombospondin-type laminin G domain and several tandem arranged epilepsy-associated repeats (EARs). A mutation in this gene is the cause of autosomal recessive deafness-98. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Dec 2012]

Variant classified as Uncertain Significance - Favor Benign. The p.Thr27Thr (c.8 1A>C ) variant in TSPEAR has not been previously reported in individuals with he aring loss and was absent from large population studies. This variant is located in the second to last base of the exon, which is part of the 5' splice region. Computational tools do not suggest an impact to splicing. However, this informat ion is not predictive enough to rule out pathogenicity. In summary, while the cl inical significance of the p.Thr27Thr variant is uncertain, these data suggest t hat it is more likely to be benign. -
