This variant has not been reported in the literature in individuals with MYLK-related disease. This variant is present in population databases (rs150392984, ExAC 0.01%). This sequence change replaces aspartic acid with tyrosine at codon 1681 of the MYLK protein (p.Asp1681Tyr). The aspartic acid residue is highly conserved and there is a large physicochemical difference between aspartic acid and tyrosine. -

The p.D1681Y variant (also known as c.5041G>T), located in coding exon 27 of the MYLK gene, results from a G to T substitution at nucleotide position 5041. The aspartic acid at codon 1681 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
