Variant summary
Our verdict is Likely pathogenic. Variant got 8 ACMG points: 8P and 0B. PVS1_StrongPM2PP5_Moderate
The NM_003242.6(TGFBR2):c.1563G>A(p.Trp521*) variant causes a stop gained change involving the alteration of a conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Likely pathogenic (★).

Familial thoracic aortic aneurysm and aortic dissection Pathogenic:1
In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This nonsense change has been observed in individual(s) with TGFBR2-related conditions (PMID: 18781618, 27879313, Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 487632). This variant is not present in population databases (ExAC no frequency). This sequence change results in a premature translational stop signal in the TGFBR2 gene (p.Trp521*). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 47 amino acids of the TGFBR2 protein. -
