The HBB c.162delT; p.Val55LeufsTer7 variant (also known as codon 53 (-T), or Val54fs when numbered from the mature protein, rs1554917935) is reported in a family affected with beta thalassemia intermedia, and is inherited in a dominant fashion in this large family (Yi 2008). This variant is also reported in ClinVar (Variation ID: 439137), but is absent from the Genome Aggregation Database, indicating it is not a common polymorphism. This variant causes a frameshift by deleting a single nucleotide, so it is predicted to result in a truncated protein or mRNA subject to nonsense-mediated decay.
