Neuropathy, hereditary sensory and autonomic, type 1C Uncertain:1
Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with SPTLC2-related disease. This sequence change replaces aspartic acid with glutamic acid at codon 344 of the SPTLC2 protein (p.Asp344Glu). The aspartic acid residue is highly conserved and there is a small physicochemical difference between aspartic acid and glutamic acid. This variant is not present in population databases (ExAC no frequency). -
SPTLC2-related disorder Uncertain:1
The SPTLC2 c.1032T>G variant is predicted to result in the amino acid substitution p.Asp344Glu. To our knowledge, this variant has not been reported in the literature or in a large population database, indicating this variant is rare. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
