The BTB domain functions as a protein-protein interaction module, which includes an ability to self-associate or to interact with non-BTB domain-containing proteins. The kelch motif typically occurs in groups of five to seven repeats, and has been found in proteins with diverse functions. Known functions of these family members include transcription regulation, ion channel tetramerization and gating, protein ubiquitination or degradation, and cytoskeleton regulation. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 402 amino acid(s) of the KBTBD13 protein. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant disrupts the p.Arg408 amino acid residue in KBTBD13. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 21109227). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. ClinVar contains an entry for this variant (Variation ID: 532998). This variant has not been reported in the literature in individuals affected with KBTBD13-related conditions.
