The BTB domain functions as a protein-protein interaction module, which includes an ability to self-associate or to interact with non-BTB domain-containing proteins. The kelch motif typically occurs in groups of five to seven repeats, and has been found in proteins with diverse functions. Known functions of these family members include transcription regulation, ion channel tetramerization and gating, protein ubiquitination or degradation, and cytoskeleton regulation. The frame shift c.477del p.Val160Ter variant in KBTBD13 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.Val160Ter variant is novel not in any individuals in gnomAD Exomes and is novel not in any individuals in 1000 Genomes. This variant has been reported to the ClinVar database as Uncertain Significance. This variant is predicted to cause loss of normal protein function through protein truncation. As loss of function in this gene is not a well known mechanism, hence for these reasons, this variant has been classified as uncertain significance VUS. -
