Genes affected
DNAH11 (HGNC:2942): (dynein axonemal heavy chain 11) This gene encodes a ciliary outer dynein arm protein and is a member of the dynein heavy chain family. It is a microtubule-dependent motor ATPase and has been reported to be involved in the movement of respiratory cilia. Mutations in this gene have been implicated in causing Kartagener Syndrome (a combination of situs inversus totalis and Primary Ciliary Dyskinesia (PCD), also called Immotile Cilia Syndrome 1 (ICS1)) and male sterility. Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Jan 11, 2021 | The p.L2698S variant (also known as c.8093T>C), located in coding exon 49 of the DNAH11 gene, results from a T to C substitution at nucleotide position 8093. The leucine at codon 2698 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |

Uncertain significance, criteria provided, single submitter | clinical testing | GeneDx | Apr 12, 2021 | Identified in the heterozygous state without a second DNAH11 variant in an individual with primary ciliary dyskinesia; this individual also harbored a single heterozygous variant in the GAS8 gene (De Jesus-Rojas et al., 2021); In silico analysis, which includes protein predictors and evolutionary conservation, supports a deleterious effect; This variant is associated with the following publications: (PMID: 33670432) - |
