To our knowledge, functional studies have not been reported for this variant. This variant has been reported in a cohort of African men diagnosed with prostate cancer (PMID: 32832836). This variant has been identified in 6/281184 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The p.K866T variant (also known as c.2597A>C), located in coding exon 4 of the MSH6 gene, results from an A to C substitution at nucleotide position 2597. The lysine at codon 866 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species.
