The p.N557D variant (also known as c.1669A>G), located in coding exon 7 of the PKP2 gene, results from an A to G substitution at nucleotide position 1669. The asparagine at codon 557 is replaced by aspartic acid, an amino acid with highly similar properties. This alteration has been reported in an arrhythmogenic disorders cohort (Marschall C et al. Cardiovasc Diagn Ther, 2019 Oct;9:S292-S298). This alteration has also been reported in a pediatric cardiomyopathy cohort (Ware SM et al. Am J Hum Genet, 2022 Feb;109:282-298). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
