The p.R1377H variant (also known as c.4130G>A), located in coding exon 4 of the PRX gene, results from a G to A substitution at nucleotide position 4130. The arginine at codon 1377 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Charcot-Marie-Tooth disease type 4 Uncertain:1
ClinVar contains an entry for this variant (Variation ID: 583179). This variant has not been reported in the literature in individuals affected with PRX-related conditions. This variant is present in population databases (rs200835105, gnomAD 0.03%). This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 1377 of the PRX protein (p.Arg1377His).
