The HBB c.22G>T (p.Glu8*) variant causes the premature termination of HBB protein synthesis. This variant has been reported to be associated with hemolytic anemia and ineffective erythropoiesis (ITHANET (http://www.ithanet.eu/)). It has also been reported in large beta thalassemia screening studies (PMID: 18096416 (2008), 33947296 (2021)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Based on the available information, this variant is classified as pathogenic. -
