Variant summary: GAA c.2092G>A (p.Ala698Thr) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 4.9e-05 in 225392 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in GAA causing Glycogen storage disease, type II, allowing no conclusion about variant significance. c.2092G>A has been observed in at least one individual affected with cramps, myalgia, and hyperCKemia (e.g., Gemelli_2021). This report does not provide unequivocal conclusions about association of the variant with Glycogen storage disease, type II. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
