The arginine at codon 1009 is replaced by tryptophan, an amino acid with dissimilar properties. This variant was detected in a reportedly healthy exome cohort (Landstrom AP et al. Circ Arrhythm Electrophysiol, 2017 Apr;10:pii: e004742) as well as in a stillbirth cohort (Sahlin E et al. PLoS ONE, 2019 Jan;14:e0210017). This alteration was also noted in a pediatric cardiomyopathy cohort (Ware SM et al. Am J Hum Genet, 2022 Feb;109:282-298). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
