The c.7G>T (p.G3W) alteration is located in exon 1 (coding exon 1) of the FAM175A gene. This alteration results from a G to T substitution at nucleotide position 7, causing the glycine (G) at amino acid position 3 to be replaced by a tryptophan (W). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

Variant summary: FAM175A c.7G>T (p.Gly3Trp) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-05 in 225418 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.7G>T has been reported in the literature in individuals affected with Sarcoma, who also carried other variants (Ballinger_2016). This report does not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 27498913). One submitter has cited clinical-significance assessments for this variant to ClinVar after 2014 and has classified the variant as uncertain significance.
