FBN1 (HGNC:3603): (fibrillin 1) This gene encodes a member of the fibrillin family of proteins. The encoded preproprotein is proteolytically processed to generate two proteins including the extracellular matrix component fibrillin-1 and the protein hormone asprosin. Fibrillin-1 is an extracellular matrix glycoprotein that serves as a structural component of calcium-binding microfibrils. These microfibrils provide force-bearing structural support in elastic and nonelastic connective tissue throughout the body. Asprosin, secreted by white adipose tissue, has been shown to regulate glucose homeostasis. Mutations in this gene are associated with Marfan syndrome and the related MASS phenotype, as well as ectopia lentis syndrome, Weill-Marchesani syndrome, Shprintzen-Goldberg syndrome and neonatal progeroid syndrome. ESE finder predicts that this variant may affect ESE site of SRp55 and SC35.However, these predictions have yet to be confirmed by functional studies. This variant was found in 21/276618 control chromosomes, predominantly observed in the European (Non-Finnish) subpopulation at a frequency of 0.000166 (21/126254). This frequency is somewhat higher than the estimated maximal expected allele frequency of a pathogenic FBN1 variant (0.0001125), suggesting this is likely a benign polymorphism found primarily in the populations of European (Non-Finnish) origin. The variant was reported in the literature as a polymorphism (Katzke 2002). In addition, clinical diagnostic laboratories classified this variant with conflicting interpretations of pathogenicity (1 likely benign / 1 uncertain significance). Taken together, this variant is classified as VUS-possibly benign. -
