Although this v ariant has been seen in the general population, its frequency is not high enough to rule out a pathogenic role. The Alanine (Ala) at position 57 is not conserve d in mammals, and 2 mammals carry a Threonine (Thr) despite high nearby amino ac id conservation, raising the possibility that this change at this position may b e tolerated.
