The Alanine (Ala) at position 57 is not conserve d in mammals, and 2 mammals carry a Threonine (Thr) despite high nearby amino ac id conservation, raising the possibility that this change at this position may b e tolerated. Additional computational prediction tools suggest that the p.Ala57T hr variant may not impact the protein, though this information is not predictive enough to rule out pathogenicity.
