Mutations in this gene are associated with Wolfram syndrome, also called DIDMOAD (Diabetes Insipidus, Diabetes Mellitus, Optic Atrophy, and Deafness), an autosomal recessive disorder. The disease affects the brain and central nervous system. Mutations in this gene can also cause autosomal dominant deafness 6 (DFNA6), also known as DFNA14 or DFNA38. Although this v ariant has been seen in the general population, its frequency is not high enough to rule out a pathogenic role. The Alanine (Ala) at position 57 is not conserve d in mammals, and 2 mammals carry a Threonine (Thr) despite high nearby amino ac id conservation, raising the possibility that this change at this position may b e tolerated. Additional computational prediction tools suggest that the p.Ala57T hr variant may not impact the protein, though this information is not predictive enough to rule out pathogenicity.
