The p.Pro2591Leu variant in DNAH11 has been reported, in the compound heterozygous state, in 1 individual with primary ciliary dyskinesia (Variation ID: 36981; PMID: 22102620), and has been identified in 0.0008% (9/1176488) of European (non-Finnish) chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP ID: rs387907258). Although this variant has been seen in the general population in a heterozygous state, its frequency is low enough to be consistent with a recessive carrier frequency. Computational prediction tools and conservation analyses do not provide strong support for or against an impact to the protein. This variant has been reported in combination with a pathogenic variant in DNAH11 in an individual affected with primary ciliary dyskinesia (PMID: 22102620). ClinVar contains an entry for this variant (Variation ID: 36982). This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with leucine at codon 2591 of the DNAH11 protein (p.Pro2591Leu). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and leucine. -
