The lysine at codon 68 is replaced by asparagine, an amino acid with similar properties. This alteration has been identified in multiple individuals with pancreatitis (Gaia E et al. Dig Dis Sci, 2002 Nov;47:2416-21; Sofia VM et al. Mol Med, 2016 Sep;22:300-309). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |

The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.204A>T has been reported in the literature as a non-informative genotype (second allele of uncertain significance or not reported) in individuals affected with Pancreatitis, Cystic Fibrosis or in settings for newborn screening (example, Sofia_2016, Gaia_2002, Handschick_2012, Bozdogan_2020). These report(s) do not provide unequivocal conclusions about association of the variant with Cystic Fibrosis. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 33572515, 12452372, 21917531, 24777605, 25735457, 27264265).
