The p.G2595V variant (also known as c.7784G>T), located in coding exon 62 of the FBN1 gene, results from a G to T substitution at nucleotide position 7784. The glycine at codon 2595 is replaced by valine, an amino acid with dissimilar properties. This alteration has been reported in a subject with features of Marfan syndrome (Lerner-Ellis JP et al. Mol Genet Metab, 2014 Jun;112:171-6). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
