Myosin-binding protein C is a myosin-associated protein found in the cross-bridge-bearing zone (C region) of A bands in striated muscle. MYBPC3 is expressed exclusively in heart muscle and is a key regulator of cardiac contraction. Mutations in this gene are a frequent cause of familial hypertrophic cardiomyopathy. Alanine (Al a) at position 109 is not conserved and possibly not present in multiple mammals , and the change to Valine (Val) was predicted to be benign using a computationa l tool clinically validated by our laboratory. This tool's benign interpretation is estimated to be correct 89% of the time (Jordan 2011). In summary, additiona l studies are needed to fully assess its clinical significance. -
