The arginine at codon 143 is replaced by leucine, an amino acid with dissimilar properties. This alteration has been reported in exome sequencing and hypertrophic cardiomyopathy (HCM) cohorts (Homburger JR et al. Proc Natl Acad Sci U S A, 2016 Jun;113:6701-6; Harper AR et al. Nat Genet, 2021 Feb;53:135-142). Other alterations affecting the same amino acid, p.R143W (c.427C>T) and p.R143Q (c.428G>A), have been reported in association with hypertrophic cardiomyopathy (HCM) (Erdmann J et al. Clin.
