This sequence change affects a donor splice site in intron 4 of the SDHB gene. RNA analysis indicates that disruption of this splice site induces altered splicing and likely results in the loss of 18 amino acid residue(s), but is expected to preserve the integrity of the reading-frame. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individuals with paraganglioma (PGL) and/or pheochromocytoma (PCC) (PMID: 16405730, 19411806, 19825962, 20540712, 21348866, 25047027). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 1326294). Studies have shown that disruption of this splice site results in the activation of a cryptic splice site in exon 4 (PMID: 16405730, 19411806). This variant disrupts a region of the SDHB protein in which other variant(s) (p.Val140Phe) have been determined to be pathogenic (PMID: 16912137, 19189136, 19802898). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic. -
