TSC2 (HGNC:12363): (TSC complex subunit 2) This gene is a tumor suppressor gene that encodes the growth inhibitory protein tuberin. Tuberin interacts with hamartin to form the TSC protein complex which functions in the control of cell growth. This TSC protein complex negatively regulates mammalian target of rapamycin complex 1 (mTORC1) signaling which is a major regulator of anabolic cell growth. The Y429X nonsense variant in the TSC2 gene is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Y429X variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). A nonsense variant at the same residue [c.1287T>G (p.Y429X)] has been previously reported as a sporadic variant in an individual with tuberous sclerosis complex (Au et al., 2007). Although this pathogenic variant has not been reported previously to our knowledge, its presence is consistent with the diagnosis of TSC in this individual. -
