The tryptophan at codon 827 is replaced by leucine, an amino acid with similar properties. Another alteration affecting the same amino acid, p.W827C (c.2481G>T), has been reported in association with hypertrophic cardiomyopathy (HCM) (Millat G et al. 2010 Eur J Med Genet Jul;53:261-7). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
