Variant summary
Our verdict is Likely benign. The variant received -2 ACMG points: 0P and 2B. BP4BS2_Supporting
The NM_000465.4(BARD1):c.2131A>G(p.Ser711Gly) variant causes a missense change involving the alteration of a conserved nucleotide. The variant allele was found at a frequency of 0.00000434 in 1,614,226 control chromosomes in the GnomAD database, including 1 homozygotes. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. S711R) has been classified as Uncertain significance.

Familial cancer of breast Uncertain:2
This sequence change replaces serine, which is neutral and polar, with glycine, which is neutral and non-polar, at codon 711 of the BARD1 protein (p.Ser711Gly). This variant is present in population databases (rs564838936, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with BARD1-related conditions. ClinVar contains an entry for this variant (Variation ID: 460738). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
not provided Uncertain:1
Not observed at significant frequency in large population cohorts (gnomAD); In silico analysis supports that this missense variant does not alter protein structure/function; Has not been previously published as pathogenic or benign to our knowledge; This variant is associated with the following publications: (PMID: 17550235, 28202063)
Hereditary cancer-predisposing syndrome Uncertain:1
The p.S711G variant (also known as c.2131A>G), located in coding exon 11 of the BARD1 gene, results from an A to G substitution at nucleotide position 2131. The serine at codon 711 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
