The p.A259V variant (also known as c.776C>T), located in coding exon 8 of the ACTN2 gene, results from a C to T substitution at nucleotide position 776. The alanine at codon 259 is replaced by valine, an amino acid with similar properties. This alteration has been reported in association with cardiomyopathy (Ware SM et al. Am J Hum Genet, 2022 Feb;109:282-298; Akinrinade O et al. J Cardiovasc Transl Res, 2023 Jul;:). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
