The p.D1240N variant (also known as c.3718G>A), located in coding exon 30 of the FBN1 gene, results from a G to A substitution at nucleotide position 3718. The aspartic acid at codon 1240 is replaced by asparagine, an amino acid with highly similar properties, and is located in the cbEGF-like #16 domain. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.3718G>A in individuals affected with Marfan Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. Four submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as uncertain significance.
