In Drosophila crumbs localizes to the stalk of the fly photoreceptor and may be a component of the molecular scaffold that controls proper development of polarity in the eye. Mutations in this gene are associated with a severe form of retinitis pigmentosa, RP12, and with Leber congenital amaurosis. Alternate splicing results in multiple transcript variants, some protein coding and some non-protein coding.[provided by RefSeq, Apr 2012]

This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 947 of the CRB1 protein (p.Glu947Lys). This variant is present in population databases (rs574162883, gnomAD 0.06%). This missense change has been observed in individuals with autosomal recessive retinitis pigmentosa (PMID: 28041643, 32581362). ClinVar contains an entry for this variant (Variation ID: 438077).
