This variant is only observed on two alleles in the Genome Aggregation Database, indicating it is not a common polymorphism. The p.D1169Y variant (also known as c.3505G>T), located in coding exon 19 of the BRIP1 gene, results from a G to T substitution at nucleotide position 3505. The aspartic acid at codon 1169 is replaced by tyrosine, an amino acid with highly dissimilar properties. This alteration was observed within 1 of 64523 individuals with a personal history of breast cancer (Easton DF et al. J Med Genet, 2016 05;53:298-309). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
