The p.P253L variant (also known as c.758C>T), located in coding exon 7 of the MRE11A gene, results from a C to T substitution at nucleotide position 758. The proline at codon 253 is replaced by leucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 253 of the MRE11 protein (p.Pro253Leu). This variant is present in population databases (rs587782051, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with MRE11-related conditions. This missense change has been observed to be homozygous or hemizygous in an individual who did not have the expected clinical features for that genetic result (Invitae).
