ABCC9 (HGNC:60): (ATP binding cassette subfamily C member 9) The protein encoded by this gene is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the MRP subfamily which is involved in multi-drug resistance. This protein is thought to form ATP-sensitive potassium channels in cardiac, skeletal, and vascular and non-vascular smooth muscle. Protein structure suggests a role as the drug-binding channel-modulating subunit of the extra-pancreatic ATP-sensitive potassium channels. Mutations in this gene are associated with cardiomyopathy dilated type 1O. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Apr 2011]

Although this variant is predicted to be deleterious to the protein, the variant spectrum of this gene has not been well characterized and it is unclear if variants of this type play a role in disease. The c.4196dupT variant has not been published as pathogenic or been reported as benign to our knowledge. The c.4196dupT variant is not observed in large population cohorts (Lek et al., 2016). This variant causes a shift in reading frame starting at codon serine 1400, changing it to a glutamine, and creating a premature stop codon at position 5 of the new reading frame, denoted p.Ser1400GlnfsX5. This variant is expected to result in either an abnormal, truncated protein product or loss of protein from this allele through nonsense-mediated mRNA decay. However, only a few loss of function variants in the PRDM16 gene have been reported in Human Gene Mutation Database in association with ABCC9-related disorders (Stenson et al., 2014), indicating that loss of function is not an established mechanism of disease for this gene. -
